She was first admitted to hospital on 2 August with flu-like symptoms but later transferred to an intensive care unit with cardiac-related problems.
The Welsh Assembly Government confirmed that the woman from Caerphilly county borough died on Saturday following further circulatory complications.
There have been more than 50 swine flu-related deaths in the UK so far.
Health Minister Edwina Hart said there was no increase in case rates in Wales.
The woman was first admitted to the Royal Gwent Hospital in Newport where she tested positive for swine flu on admission and received a course of antivirals.
She was later transferred to the intensive care unit at the University Hospital of Wales in Cardiff.
